TRANSCEND-T2D-1: glycemic control in phase 3

Study question & design
This double-blind phase 3 trial randomized 537 adults with type 2 diabetes inadequately controlled by diet and exercise to retatrutide or placebo.
Reported findings
All three retatrutide groups showed greater reductions in glycated hemoglobin than placebo under the reported treatment-regimen analysis.
Limits of interpretation
The endpoint was glycemic control in a selected diabetes population. It does not by itself establish long-term cardiovascular benefit, general safety or equivalence of other product preparations.

Study question & design
Investigators reanalyzed blood biomarkers from two phase 2 trials in adults with obesity or overweight, with or without type 2 diabetes.
Reported findings
Several atherogenic lipoprotein measures decreased. Selected inflammatory markers improved in the study without diabetes but not in the diabetes study.
Limits of interpretation
This was a post hoc analysis of surrogate markers, not a trial establishing fewer heart attacks or strokes. Results should retain their population-specific differences.
